New Research on Hearing Loss Reversal

Researchers might have opened the door to reversing some types of hearing loss, and it’s fascinating. Recent studies indicate that hearing loss linked to certain genetic mutations can, in fact, be reversed through a combination of common supplements and the erectile dysfunction medication Viagra (sildenafil).

The investigation has pinpointed mutations in a gene called CPD, which generates an enzyme known as carboxypeptidase D. This was observed in three distinct families from Turkey, where several members were born deaf.

According to the National Institutes of Health, all these families exhibit hereditary sensorineural hearing loss. This condition is attributed to damage sustained by the delicate hair cells in the inner ear, which are responsible for transforming sound vibrations into electrical signals that the brain interprets.

To delve deeper into this relationship, researchers analyzed genetic data from both affected and unaffected members of these families and extended their research to different groups.

Insights from Clinical Tests

In a UK genome sequencing cohort, researchers discovered that mutations in the CPD gene were present in individuals with hearing loss, as noted by Dr. Mustafa Tekin, a professor of human genetics at the University of Miami Miller School of Medicine.

By turning off the CPD gene in human cells during clinical tests, the researchers found reduced levels of three substances: arginine (an amino acid), cyclic guanosine monophosphate (cGMP, a vital signaling molecule), and nitric oxide, which serves crucial signaling roles in the nervous system.

Interestingly, while the initial research was conducted on humans, experiments were also carried out on mice and fruit flies. Low levels of nitric oxide and cGMP in these animals led to significant stress on the hair cells in their inner ears, ultimately resulting in cell death.

When researchers administered arginine to human cells, nitric oxide levels returned to standard, which helped reduce cell mortality. Moreover, Viagra seemed to reverse some of the detriments brought about by CPD mutations, according to Tekin.

This research suggests that a deficiency in nitric oxide within the cochlea, the area in the inner ear responsible for converting sound into nerve signals, could be a major contributor to hearing loss. The CPD gene seems to regulate the maintenance of arginine and nitric oxide levels in hair cells, which is essential for effective signal transmission and protection against noise damage.

Dr. Tekin stressed, “Nitric oxide is critical for multiple tissues, and a careful balance is necessary.” However, he clarified that they aren’t promoting Viagra as a treatment for hearing loss.

Expert Caution

Mirroring this sentiment, other experts have voiced concerns regarding potential side effects of Viagra. Dr. Noushin Perhizkar, an otolaryngologist from the San Francisco Bay Area, highlighted that the drug has been associated with rare instances of sudden hearing loss and irreversible tinnitus, emphasizing the need for caution.

She stated, “At this point, it’s not appropriate or safe for the general public to utilize Viagra for hearing loss.”

Although studies indicate that hearing loss is often temporary and resolves when the medication is discontinued, the FDA has included warning labels on Viagra and similar medications about the risk of sudden hearing loss following rare reports from users.

However, experts remain uncertain if this side effect is directly linked to PDE5 inhibitors since affected individuals may have other underlying health issues or be taking different medications that could contribute to hearing loss.

Limitations of the Research

The study was conducted on a very small and genetically defined group of patients, so its findings may not extend to the broader population. Dr. Perhizkar cautioned that it is not yet safe for the general public to consider Viagra as a remedy for hearing loss.

As it stands, the research primarily focused on restoring hearing loss in animal models, indicating a need for further studies to determine the effectiveness in humans. Dr. Tekin also emphasized the importance of genetic testing to identify the underlying causes of hearing loss, mentioning that scientists have found over 200 genes associated with this condition. However, most available genetic screening tests may only concentrate on the most frequently occurring mutations.
